Design Notes

The toehold switch should form a hairpin structure, where the base of the bottom stem structure. It should the basic design of the second generation toehold switch of Pardee et al. 2016.1 The minimum free energy of the complex of toehold switch annealed to the miRNA should also be lower compared to the MFE of the toehold switch and miRNA alone in the solution.
